The Role of ACE2 Gene polymorphisms in men Hypertension patients and COVID 19

Abstract

Coronavirus disease 19 (COVID-19) is severe acute respiratory syndrome that caused by (SARS-CoV-2) which causing human health problems. Aim of this study is determine the relationship between genetic variants of ACE2 gene receptor with COVID 19- and Hypertension disease comparing with healthy control group using ARMS-PCR technique, and Estimation of ACE2 levels in serum samples of patient group and control group. Rs4341 polymorphism of enzyme2 (ACE2) gene, key regulator of the renin-aldosterone-angiotensin system (RAAS), This study include 100 samples were collected from males infected with and without COVID-19 from different areas of the governorates of Babylon, Their ages ranged from (40–63) years, divided in to fourth group, first group include 25 COVID-19 with hypertensive patients, second group 25 COVID-19 without hypertensive patients, third group 25 hypertensive patients and fourth group 25 healthy people. The results of this study show frequencies distribution of ACE2 (rs4341) genotypes were GG genotype type and allele G which were clearly significant in serum levels of ACE2 in COVID-19 patients with hypertension vs COVID-19 patients without hypertension, and healthy control group according to ACE2 (rs4341) genotypes and alleles were (p<0.001). So the ACE2 (rs4341) GG genotype distribution showed a highly significant difference in the gene allele GG distribution in COVID-19 patients with hypertension compared to COVID-19 patients without hypertension and normal control (P=0.030). But in Covid-19 cases without hypertension, the present results show non-significant increased the serum ACE2 levels in GG genotype (P=0.372).  Our work suggests that G-containing genotypes of rs4341 and rs4343 are risk factors for COVID-19 development. ACE2 polymorphisms in COVID-19 cases with hypertension is indicator for high severity in COVID-19 cases with hypertension
